I have a 2000 Rodeo and when I accelerate - when it shifts from to 2nd gear, the car jerks back and makes a sort of hiccup (screeching) noise. I have about 61,000 miles on the car. Recently had a transmission service and it got rid of the problem for about a month. Now it's back. Any ideas of what this is???

Run your transmission selector through the gears and watch the dash selector. Do they match up? If you have it in drive, and you pull back a little (not enough to go into 3rd) does it drop into third? If so clean up you range selector connections (see the faq's).
Also, isuzus do funky things with weak batteries, alts and dirty ground points (the key one is the ground to the alt bracket). Who flushed it? Isuzu?

Do the 97 model have a sealed tranny? can you change the trans fluid on it, mine shifts kind of hard

Yes it is sealed (meaning there is no dipstick to check it) and yes you still need to change the fluid every 30-50k.
