how was it running before? did you have a timing belt job done?
you should be able to turn it over completely all the way by hand. i'd start ruling things out. check and make sure all the timing marks are lined up. if they're good i'd try turn it over it might be the tranny locking it up. have someone turn it over while you listen around a couple different areas to see if you can hear any clicking or rubbing or anything like that.
