Generally, there is no relation to the valve stem and the tire. The tire can be located anywhere in relation to the tire.
As for mounting a tire, its best to take it to a tire shop, since this is a safety related issue and not for a beginner. It is easy to damage the tire or rim, which would make either component dangerous. Also, the first time you inflate a tire on a rim (to seat the bead) it is possible to have a tire explosion that can cause injury or death if the tire is not installed correctly.
