do you have plenty of coolant? it sounds like youre truck is overheating.

~acts up during the day after being run for 34 miles

those are all symptoms of overheating/low coolant
what I would do is get a timing gun and hook it up to cylinder 1 and see if its getting any power at all if it is, change the plug again. if it still misfires, test the injector.
