Sure, it can be done but would seriosly weaken the structure.
The car in the picture has a roll cage in it to make up for the loss of the roof structure.
Without the roof, the car would want to twist over the bumps and would bend in the middle a bit over bumps and heavy acceleration/braking.
This would prevent you from getting a good fit for the targa panel.
If you do not want a roll cage, you can add frame rails to the bottom of the car. This is done with custom made convertables.
I would suggest just add a sunroof on its own (if you do not already have one) and save the structural hassles.
