Based on some of your other posts, I believe you have a V8. In that case, cylinders 3 and 5 are not near the back of the engine where they're sometimes prone to moisture from water running off the windshield and onto the valve covers.
If the coil has actually failed, you will need to replace it, not just the boots and contact springs. To verify whether or not the coil is faulty, you could try relocating it to another cylinder and see if the misfire code follows the coil or the cylinder. You should try drying the connector and determining how the moisture got in the connector to begin with. Are you sure it's moisture in the connector and not dielectric grease or similar?

I think there was a recall on the Cowell,it was diverting water onto
buss lines, the water rolls in behind COP covers.
Use some Silicone sealent behind COP covers, where the electrical ignition wires poke in.
Try using a hair dryer and Alcohol to dry COP's + Spark plug boss tubes.
The Heat of Motor will dry up water in Spark Plug boss tubes, but will leave evidence
of a rust residue on your COP boots.

I ended up using a rubber 4'' baseboard molding from Home Depot!
You can make your own Rain diverter.
