Simple. I spray everything I can, and I brush whatever is impractical to spray. What can and can't be sprayed varies by car, kit, and builder's skills.

Since I don't have an air brush yet, I do alot of brush painting and masking. I cut up my sprues leaving number then spray what I can. I've found that acrylics give me better control and less brush lines than enamels, but you must prime!

do you guys spray before you punch it out of the plastic holders? or do you glue everything 1st?

I pour the paint into the box and shake.... :)
It depends on the part and what you prefer.
Personally, I remove 99.9% of parts from the sprue, clean up mold lines with sand sticks and/or hobby knife, stick the parts onto bamboo skewers with double-sided tape and then prime and spray everything with my airbrush.

By punch I hope you don't mean you break the parts off the sprue, use a pair of cutters or Xacto knife to carefully remove parts from the sprue and reduce the risk of damaging parts.
Paint as much as you can before assembling with glue - if you've got paint on a part where it's got to be glued, carefully remove the paint with your Xacto.
