Can anyone tell me if the bypass hole of the thermistat is to be at a certain position when installing (20001 Buick 3.8 ),the thermistat is offset thats why I'm asking.

Good to know Buick is still around in 20001! I bet they still use the 3800 too!
Just make sure its tight. fill the radiator with coolant, and the overflow tank to full cold. Do not cap the radiator. Start the engine and let it warm up, so that the thermostat will open. Watch the level of the coolant in the radiator, and fill as it lowers, and be ready to cap it quickly. Then just fill the overflow tank to full cold. Keep an eye on the temp sensor in the car(should be at 195) and keep the coolant tank at full hot once its warmed up.

Thanks T man but my question was as posted ,anyway the replacement was not offset and didn't have a bypass hole but works very good, have a good set of Holidays.

I noticed that with the last one I replaced on my 1998. I put the hole at the uppermost part since it's to allow air to easily move out into the hose. Air would slowly make it around the moveable vane when closed but the hole allows it to go faster making filling easier.
