Can I use Plus on the RX300 or I must use Premium?

I think you can use regular. New cars these days are designed that way. John Cambece, Peabody, MA

The owners manual says you can use either. I tried both and I did not get enough MPG increase to offset the extra cost. If there was increased performance, I didn't notice that either.

Maybe you should try both and see what works best for you.
