I have a 2004 Sierra extended cab. It has developed a squeak that I believe to be in the latch area on the upper portion of the cab. It squeaks every time a bump in the road is hit. Does anyone have any knowledge of what may be causing this problem? Thanks for your time.

Check for rubs in the paint in the general area. More than likely it will be on the inside/bottom where the door has been rubbing on the body. If so, take a look at the hinges and verify tightness of the welds and bolts. If they are off, don't screw with them...if you have that new a truck, I can only assume that it is still under warranty. Point it out to the lackies at the dealer.
If there is no rub, then try a Lithium-based grease from any auto parts supply. They do sell a spray version, but my personal pref is the hands-on, gettin' dirty, old school small bowl. Get it up in the lock mechanism on both the door and the body.
If that doesn't work, then start searching for rubs under the door panel. You may wanna check with your dealer to verify body warranty BS before diggin in.
Good Luck
