V6 4.3L plenum gasket?

Is there any tips/tricks/problems associated with replacing the plenum gasket on the 4.3L vortec engine?

Nope, take off the old and replace with the new.
Be ready for seeing leaks if you are taking off the upper plenum.
Passenger side washed clean = Injector shot.
Driver's side clean = internal fuel lines shot.
Do not ignore them and just replace the gasket.

Also, make sure you torque down the gasket to the recommended level and do not over toque them or they will snap and you will then be in real trouble.

do you use gray ATV (high torque)? A thin layer applied to both sides of the gasket (just like other gaskets)?

Nono.
You just lay it down with the green sealant bead facing up.
Nothing is needed for install. Use no other sealants. The gasket comes with its own already applied.

Actually I am removing the plenum cover in order to assess the CPI and other systems. This truck was only getting 8mpg. So I'm thinking major fuel injection problems.
Is it possible that the FPR in the CPI is the cause of leaks on the passenger side? That is a much cheaper repair. (~$60) Or is it better to just go ahead and replace the whole CPI (~$250)?

Do you know if Lendertech replacement pressure regulator (CSFI) is worth the trouble?

Don't just replace the FRP on the side of the injector, replace the whole thing.
The cheapest place to get the injector is off of ebay.

Also, It is best to replace the nut kit while you have everything out.
The nut kit is prone to leaks as well seeing it was made of an improper platic that gets too brittle and breaks.
