My amp has Variable Low-pass 45Hz to 5kHz
Variable high-pass with INTELLi Q 10Hz to 10kHz.
How do I know where to set it?

It is powering 2 kicker comp vr 12"s they are dvc 4 ohm. The frequency response is 25-500

set it to lowpass, somewhere around 70-80hz

There's no real magic spot to put it, around 80hz for subs is just a guideline. Do you know what a lowpass filter does? If not then you might want to check out www.bcae1.com. If so I'll just go on. You basically want to set the lowpass as low as you can while still getting good frequency coverage. Your mids have a natural rolloff when the frequency drops low enough, you want to set your sub's lowpass high enough where it can pick up where the mids leave off, but not too high where the bass becomes directional and you can hear it coming from behind you. 80hz is just sort of a happy medium.
