2009 Ford Escape 2.5L misfire...help please

I am working on a 2009 ford escape 2.5 L non-hybrid and am getting a misfire code on cylinder two. Replaced the coil and spark plug but engine continues to misfire after getting up to 40mph. Below that or idle it runs perfect. I am getting spark and have proper voltage to the coil. Does anyone have any ideas what else the issue could be?
