If it's an external square fitting, big vice grips.
If it's an internal square or torx fitting, big "easy out".

the use of a rachet extension with a rachet should do it ..
if the hole is rounded get an easy out kit with various sizes and see if it will remove it ..use some heat with a small torch to get the plug hot ..
lastly use a 1/4 in drill hole on the centre of plug then use the easy out..
you must have a new plug first..
torque on the plug should be 15-20 ft lbs !

Big vise-grips. Then get a new plug.
Or just take out all the cover perimeter bolts, take off the entire cover, let a machine shop fix it, or, if cheaper, just buy a replacement cover and plug.

The fill plug is a female, 3/8" square plug. A 3/8" ratchet wrench end fits into it. It is on the side of the diff housing, not the removable back. If it got rounded off, it must be seriously seized which is odd because of the diff lube that is usually in the threads.
You'll need a big easy out, some PB Blaster and/or some freeze rust breaker spray. Alternatively, you could try to cut a slot in the head, but you'll need a LARGE screwdriver or pry bar.
Good luck & let us know how it turns out!
