Are you driving it in 4W on dry highway?
Auto Matic Transfer case with auto button on shift control panel?
Any lights 4W lights or service sone lights blinking or on?
How many miles and has transfer case fluid ever been serviced?
You.
May need to get it on a transfer case transmission capable scanner and get codes and see what is going on.
Post back any codes for the engine or ABS or tarnsmission or transfer case.
Also check all power supply to it and fuses and wiring.

I should go into more detai, The Blazer drives down the road with no problems until going down a hill and it shift out of 2 wheel drive or 4 HI but will shift into 4 Low with no problems. The only way to get it to shift back into 2 whell or 4 HI is to come to a stop and place the trans into nuteral and the transfercase into 4 LO then into 4 HI OR 2 wheel drive it slips right back in but only if you do it that way. I have replaced the contoler on the transfercase and the box down by the passengers feet. I have been driving the Blazer only in 2 wheel drive with the only time changing is to place it in 4Low to shift it back into 2 wheel. I have not had any lights blink and it is a three butten system.

You can check the 3 button transfer case for codes by grounding pin 13 and watching the shift button light flashes.
Count the flashes and post back.
Has the shift button switch been checked or tested?
Have you checked all wiring for any shorts or bad spots?
Start it up and get it in two wheel and then unplug shift button switch and see what it does with switch unpluged.
