Yeah try that first. The key should turn 90 degrees in either direction to lock/unlock, and pushing the key button should open the trunk (just in case you're not doing this right).
If the cylinder was changed, a locksmith won't be able to help. You'll have to go to a BMW dealer. Fortunately, the parts counter sells a lock cylinder repair kit, and they can code the lock to your key. At least that is the case with the driver & passenger door locks, I can't imagine that they don't do it for the trunk. This should cost you under $100, that's what my dealer told me about fixing my passenger door.
