I'm not a Toyota expert but some things that have to be considered on any conversion are are as follows.
1. You mentioned engine, tranny and ECU.
2. You need to know if the frame and motor mounts will fit a V6 or will some fabricationg, cutting and welding be needed.
3. Radiators and water hoses are probably different.
4. Wiring harness in the engine bay is probably different.
5. When you get an engine try to get it complete with alternator, starter and exhaust manifolds.
6. Air cleaner and air inlet boxes are probably different.
7. Exhaust systems are different.
8. I suspect the differential drive ratios are different but that may not be a problem.
9. Front springs for the V6 are probably stiffer than those in the I4.
10. As in any engine conversion you can plan and budget but almost everyone runs over their budget. Just be prepared.
11. If your state requires emissions inspections you may want to talk to someone familiar with emission requirements. You may find that changing engines will prevent the truck from ever passing an OBD II type inspection that many localities require due to the vehicle not matching the computer printout, just a thought.
12. This is a short list, it will get longer as you plan. I have no idea what the cost would be. I do hope some of the things I've mentioned will help.
